摘要
采用流式细胞仪(Becton Dickinson FACs Calibur)测量DNA相对含量和染色体计数2种方法,对随机选出的黄颡鱼鱼苗样本进行三倍体个体的鉴定和筛选。第1种方法是采取黄颡鱼血样,经PI染色,用流式细胞仪测定其倍性,结果14尾中有8尾黄颡鱼的DNA相对含量约是对照样本(二倍体)的1.5倍,证明为三倍体个体。染色体计数采用体内注射PHA短期培养法,经鉴定,相同样本中有8尾是三倍体,此结果验证了用流式细胞仪所测定的结果。通过比较得出,在多倍体育种工作中,尤其是诱导率不高的情况下,采集血样用流式细胞仪测定鱼体倍性是一种准确、迅速的方法,同时能够避免杀死鱼体,可以用来批量筛选多倍体鱼苗。
In this study,two identification methods were used on polysomaty analysis of yellow catfish (Pelteobagrus fulvidraco). In the first method,we took blood samples from the tail vein, and after PI staining we detected nuclear DNA relative quantity by flow cytometry in 14 fish. The results showed that DNA relative quantity of 8 fish were 1.5 times larger than normal diploid and proved that they were triptoid. The chromosome number of the same sample was also determined by the PHA colchicine hypotonic-air drying technique. Eight fish were found triploid, the same results as which by flow cytometry. By comparison of these two identification methods,we found that in the research of polyploid breeding, especially under the condition that the induction ratio of polyploid is not high, polysomaty analysis by flow cytometry is an efficient,accurate and rapid way, and it can avoid killing fish and can choose polyploid fry in batch.
