摘要
目的研究脐血源性T淋巴细胞在小鼠体内的存活、浸润情况,为深入探讨脐血源性T淋巴细胞过继性输注治疗白血病提供实验和理论依据。方法24只红白血病小鼠随机分为对照组和试验组。将分离制备的脐血源性T淋巴细胞标记后经尾静脉移植入白血病小鼠体内,采用免疫荧光、免疫组织化学等方法观察脐血源性T淋巴细胞在小鼠体内的存活时间及浸润状况。结果①移植后第3、7、14天在实验组小鼠外周血、肝和脾组织中均可检测到人CD3+细胞和Brdu+细胞;②实验组小鼠肝、脾组织内均见CD25+细胞散在浸润。结论脐血源性T淋巴细胞能在白血病小鼠体内存活并趋化至白血病细胞浸润部位。
Objective To study the survival and infiltrated condition of cord blood derived-T lymphocytes (CBTLs) in mice, to provide the experimental and theoretical basis for study on treated leukemia by cord blood-derived T cells adoptive infusion. Methods 24 erythroleukemic mice were randomly divided into two groups, the control group (CG) and the experimental group (EG). Separated and prepared cord blood derived-T lymphocytes, and transplanted it into erythroleukemic mice via the tail vein injection after signed, in order to observe the survival time and infiltrate condition of the cord blood-derived T cells in mouse by immunofluorescence (IMF), immunohistochemistry (IMC).Results①Human CD3^+ cells and Brdu^+ cells were detected in peripheral blood, liver and spleen of experimental group mice at 3st, 7st and 14st day after cord blood derived-T lymphocytes transplanting;②CD25^+ cells could be detected in liver and spleen of erythroleukemic mice in experimental group. Conclusion The Cord blood-derived T lymphocytes could be activated in vivo ,and transferred to the site of leukemia, and educed relevant function, have some therapeutic action for leukemia.
