长期吸入低浓度七氟烷对小鼠生殖功能的影响 被引量:4

Effects of long-term exposure to low-level sevoflurane on reproductive function in mice

摘要 目的评价长期吸入低浓度七氟烷对小鼠生殖功能的影响。方法ICR雄性小鼠40只,日龄60d,体重20~25g,随机分为4组(n=10):对照组(C组);S1-3组分别吸入0.003%、0.01%和0.03%七氟烷,每天2h,每周连续5d,持续8周后处死动物,取睾丸和附睾,测定精子活动率、精子数量、精子畸变率、睾丸组织总乳酸脱氢酶(LDH)及乳酸脱氢酶同工酶-X(LDH-X)的活性,并观察睾丸组织超微结构。结果与C组比较,S3组精子活动率降低、畸变率升高、睾丸组织LDH-X活性降低(P〈0.05),S1,2组各指标差异无统计学意义(P〉0.05),仅S3组睾丸组织出现病理学改变。结论长期吸入0.03%七氟烷可导致雄性小鼠生殖功能异常,而长期吸入≤0.01%七氟烷对生殖功能未见影响。 Objective To investigate the effects of long-term exposure to low-level sevoflurane on reproductive function in mice. Method Forty male ICR mice, aged 60 d, weighing 20-25 g, were randomly divided into 4 groups (n = 10 each): control group received no sevoflurane (C); group Sj_3 were exposed to 0.003% , 0.01% and 0.03% sevoflurane 2 h per day for 5 consecutive days per week for 8 weeks respectively. The mice were then sacrificed at the end of the 8 weeks. The testes and epididymis were removed and sampled for determination of the activities of total lactic dehydrogenase (LDH) and lactic dehydrogenase- X ( LDH- X ), and the motility rate, amount, and aberration rate of sperm. Testicular ultrastrueture were observed by transmission electron microscopy. Results The sperm motility rate were significantly lower, the sperm aberration rate higher and the activity of LDH- X lower in group S3 than in group C ( P 〈 0.05), but there was no significant difference in the above parameters between group Sj and group S2 ( P 〉 0.05). The pathology changed of testes occurred only in group S3 among the 3 groups. Conclusion Long-term exposure to 0.03% sevoflurane can result in the abnormality of the reproductive function in male mice but exposure to 40.01% sevoflurane dose not.
