摘要
后发性白内障主要是由于术后残留的晶体上皮细胞移行到后囊并增殖所致。用培养的新生大鼠晶体上皮细胞实验,以结晶紫染色和~3H-TdR掺入作指标,表明甘糖酯(0.2~0.8mg/ml)对细胞增殖有明显抑制作用,且比同浓度肝素作用略强。提示甘糖酯对防治后发性白内障可能有效。
Delayed opacification of the posterior capsule is the most common cause of decreased visual acuity after extracapsular cataract extraction. It has been shown to result from migration and proliferation of residual lens epithelial capsule. The effect of proglylene glucol mannate sulfate (PGMS, a polysaccharide sulfate) on the proliferation of cultured newborn rat LECs was investigated with cell culture method. LECs were incubated in modified Medium 199 at 37C in 5%CO2,and the first or second passage cells were used. The cells were exposed to 0. 2,0. 4,0. 8 mg/ml concentrations of PGMS or heparin,compared with normal control,for 72h to measure their proliferation rates by crystal violet staining and colori-metric analysis ,and 24h to account for an inhibion of 3H-TdR incorporation. The results imply that PGMS can obviously inhibit the proliferation of rat LECs in vitro(P<0. 05). PGMS, capable of inhibiting lens epithelial cells growth,if free of unacceptable toxic effects on other cell populations,might be used to prevent lens capsule opacification.
