摘要
目的:探讨白细胞介素-1α-889(interleukin-1α-889,IL-1α-889)、白细胞介素-1β-511(interleukin-1β-511,IL-1β-511)基因型与慢性牙周炎(chronic periodontitis,CP)、冠心病(coronary heartdisease,CHD)易感性的关系。方法:收集中重度CP 44例、CHD 41例、中重度CP并CHD 40例和同族健康对照49例个体的颊黏膜拭子,提取DNA,用聚合酶反应-限制性片段长度多态性法(polymerase chain reaction-restrictionfragment length polymorphism,PCR-PFLP)对IL-1α-889/NcoI,IL-1β-511/AvaI位点的基因型进行检测,比较各基因型在4组间检出率的差别。结果:①IL-1α-889基因型在4组间的分布无统计学差异。②IL-1β-511等位基因T在CHD组中的检出率显著高于健康组,而在中重度CP与健康组、中重度CP并CHD与健康组间的分布差异无统计学意义。结论:IL-1β-511等位基因T可能是冠心病的一个遗传学标志。
AIM: To investigate the interleukin -1α -889,interleukin- 1β-511 (IL- 1α -889,IL- 1β-511 )genotype and their association with the susceptibility of chronic periodontitis and coronary heart disease. METHODS: DNA was obtained from buccal swabs of 44 subjects with moderate to severe chronic periodontitis,41 subjects with coronary heart disease,40 subjects with moderate to severe chronic periodontitis and coronary heart disease and 49 matched healthy control individuals. Genotypes of IL- 1α -889/Ncoi and IL- 1β -511/AvaI were analyzed by PCR - PFLP method. Then the differences in distribution of genotype were compared. RESULTS : (1)There was no statistical differences about the distribution of IL - 1 α - 889 genotype among the four groups. (2) A significant statistical difference about distribution of IL - 1 β -511 genotype was found among the four groups. A more significant over- representation of IL- 1 β -511 allele 2 was found in CHD group than the healthy control individuals, and there was no statistical difference. CONCLUSION : This resuh suggests that the allele of IL - 1 β - 511 may be associated with CHD susceptibility.
