核因子-κB反义RNA对自发性高血压大鼠血管平滑肌细胞增殖的作用

摘要 目的:探讨核因子(NF)-κB反义RNA重组腺病毒预处理对凝血酶诱导培养的自发性高血压大鼠(SHR)血管平滑肌细胞(VSMCs)增殖的影响。方法:分别用表达NF-κB反义RNA的50多重感染(MOI)重组腺病毒和100μmol/L吡咯烷二硫代氨基甲酸(PDTC,NF-κB特异性阻断剂)预处理VSMCs 48h和1h,采用WST-1试剂和氚胸腺嘧啶核苷(3H-TdR)掺入率测定0.5 U/ml凝血酶诱导的VSMCs细胞增殖率。结果:与PDTC一样,表达NF-κB反义RNA的重组腺病毒预处理明显抑制凝血酶诱导VSMCs的增殖(P<0.01),抑制率>30%。结论:NF-κB反义RNA与NF-κB特异性阻断剂对VSMC的增殖具有相同的抑制作用,其抑制率超过30%。 Objective: To investigate the effect of antisense RNA of NF-κB on proliferation of vascular smooth muscle cells (VSMCs) in spontaneously hypertensive rats (SHR) promoted by thrombin. Methods:After pretreated with the 50MOI recombinant adenovirus expressing antisense RNA of NF-κB for 48h and 100μmol/L PDTC (NF-κB special inhibitor) for lh, VSMCs proliferation induced by 0. 5U/ml thrombin was investigated. The proliferation rate of VSMCs was determined with both WST-- 1 metabolic activity and ^3 H--TdR incorporation efficiency. Results:The pretreatment of recombinant adenovirus expressing NF-κ B antisense RNA obviously inhibit VSMCs proliferation induced by thrombin (P〈0. 01) at the rate of 〉30%, which was similar to the inhibition of VSMCs proliferation by NF-κ B special inhibitor PDTC with no statistical difference between two group(P〉0.05). Conclusion: The recombinant adenovirus expressing NF-κB antisense RNA may effectively inhibit VSMCs proliferation induced by thrombin,is similar to the NF-κB special inhibitor,may be applied to successive basic research and clinical interference treatment.
