摘要
目的观察重组人白细胞介素-1β(rhIL-1β)对人外周血淋巴细胞(PBLC)增殖及产生白细胞介素-2(IL-2)和白细胞介素-10(IL-10)的影响及芍药苷(Pae,抗炎免疫中药)对PBLC的作用。方法用密度梯度离心法,分离人PBLC;用MTT法检测PBLC的增殖反应;用放射免疫法检测PBLC培养的上清液中IL-2水平;用酶联免疫吸附法检测PBLC培养的上清液中IL-10水平。结果体外给予rhIL-1β(1×10-2、0.1、1.0、10.0、100.0μg.L-1)可不同程度促进人PBLC增殖和IL-2产生;但可抑制IL-10产生。体外给予1μg.L-1rhIL-1β(3、6、12、24、48 h),可使人PBLC产生IL-2水平升高和IL-10水平降低;不同浓度(1×10-9、1×10-8、1×10-7、1×10-6、1×10-5mol.L-1)Pae体外可不同程度抑制1μg.L-1rhIL-1β对人PBLC促增殖反应和IL-2产生;但可促进IL-10产生。结论rhIL-1β体外,可促进人PBLC的增殖,升高IL-2水平;而降低IL-10水平。Pae体外可逆转rhIL-1β对其增殖及产生IL-2和IL-10的影响。
Objective To investigate the effects of recombinant human interleukin-1β (rhIL-1β) on proliferation and producing interleukin-2 (IL-2) and interleukin-10 (interleukin-10, IL-10) of Peripheral blood lymphocyte(PBLC) from human and the effects of paeoniflorin (Pae). Methods PBLC was seperated by density gradient centrifugation. The proliferative response of PBLC was measured by MTT assay. The level of IL-2 and IL-10 in PBLC culture supernatants were detected by radioimmunoassay ( RIA ) and enzyme-linked immunsorbent assay (ELISA) respectively. Results The proliferation and IL-2 production of human PBLC treated by rhIL-1β( 1 × 10^-2 ,0. 1,1.0,10. 0,100.0 μg ·L^-1 ) were promoted, and IL-10 production was inhibited. 1 μg· L^-1 rbIL-1β (3,6,12,24,48 h)could increase the level of IL-2 and decrease the level of IL-IO produced by PBLC. Pae( 1 × 10 ^-9,1 × 10^ -8 ,1 × 10 ^-7,1 × 10^ -6, 1 × 10^ -5 mol · L^-1 ) in vitro inhibited the proliferation and IL-2 production of PBLC promoted by 1 μg ·L^-1 rhIL-1β, and increased the level of IL-10. Conclusion rhIL-1β could promote the proliferation of PBLC from human, increase the level of IL-2 and decreas the level of IL-10. Pae in vitro could reverse the effects of rhIL-1β on proliferation and producing IL-2 and IL-10 of PBLC.
