摘要
目的:对威灵仙总皂苷(TSRC)抗类风湿关节炎的作用机制进行研究。方法:利用刀豆蛋白A(ConA)诱导小鼠T淋巴细胞增殖模型;利用大鼠IL-1β、IL-2、TNF-α定量酶联检测试剂盒测定原发性佐剂性关节炎大鼠血清中IL-1β、IL-2、TNF-α的含量;利用氢氧化钾-甲醇溶液法测定原发性佐剂性关节炎大鼠肿胀足前列腺素E2(PEG2)的含量。结果:TSRC能抑制ConA诱导小鼠T淋巴细胞的增殖;降低大鼠佐剂性关节炎细胞因子(IL-1β、IL-2、TNF-α)的含量;降低佐剂性关节炎大鼠炎性肿胀足PGE2的含量。结论:TSRC可能是通过抑制T淋巴细胞的过度增殖、抑制细胞因子和PEG2来发挥抗类风湿性关节炎作用。
Aim: To study the action mechanism of total saponinis of Radix Clematidis (TSRC) on rheumatoid arthritis ( RA). Methods: The proliferation of ConA-induced T lymphocyte model was observed to find out whether TSRC had some effect on the proliferation of T lymphocytes; The serum concentrations of interleukin-1 (IL-1β), interleukin-2 (IL-2) and TNF-α in adjuvant-induced primary arthritis rats were measured by the enzyme-linked immunoadsorbent assay (ELISA) kits. And the concentrations of PGE2 in the swollen feet of adjuvant-induced primary arthritis rats were measured by the method of potassium hydroxide-methanol. Results: It was found that TSRC inhibited the proliferation of ConA-induced T lymphocytes in mice and decreased the concentrations of both the cell factor and PGE2 in the adjuvant-induced primary arthritis rats. Conclusion: The anti-RA effect of TSRC might attributed to the inhibition on the proliferation of T lymphocytes in mice and the attenuations in the concen- tration of both cell factors and PGE2 in the adjuvant-induced primary arthritis rats.
