CO2气腹对胃癌细胞SGC-7901裸鼠腹腔增殖和侵袭能力的影响 被引量:4

The influence of CO_2 pneumoperitoneum on the proliferation and metastasis of gastric cancer cells in abdominal cavity of nude mice

摘要 目的通过体内模拟腹腔镜气腹环境,探讨CO2气腹对裸鼠腹腔胃癌细胞增殖和侵袭转移能力的影响。方法裸鼠随机分成3组:对照组(A组),行开腹探查;5mmHg(1mmHg=0.133kPa)CO2气腹组(B组);8mmHgCO2气腹组(C组);每组9只。人胃癌细胞SGC-7901裸鼠腹腔注射后建立CO2气腹胃癌细胞裸鼠腹腔种植模型,气腹持续1h。建模后每周秤体质量,观察各组裸鼠生长情况和腹腔成瘤情况。28d后处死解剖裸鼠,记录腹水量、腹腔各脏器、穿刺口和腹壁切口成瘤情况和肿瘤数,测量肿瘤最大径、总重量,并检测胃癌种植瘤组织中乙酰肝素酶(Hpa)mRNA的表达。结果各组裸鼠成瘤率100%。各组裸鼠体质量、腹腔不同脏器成瘤率、腹水量差异均无统计学意义(P>0.05)。肿瘤结节数:A组(7.4±2.5)个,B组(7.3±2.4)个,C组(7.5±2.1)个。肿瘤最大径:A组(7.0±2.2)mm,B组(6.9±2.0)mm,C组(6.8±1.9)mm;肿瘤重量:A组(0.99±0.30)g,B组(0.87±0.20)g,C组(0.91±0.13)g。HpamRNA表达指数:A组0.41±0.07,B组0.45±0.06,C组0.43±0.09,各组间差异均无统计学意义(P>0.05)。结论在5mmHg和8mmHg气腹压力下,CO2气腹不促进胃癌细胞裸鼠腹腔内增殖和侵袭转移,也未明显影响胃癌细胞成瘤组织HpamRNA的表达。 Objective To observe the influence of CO2 pnemumoperitoneum models on the growth and metastasis of seeded SGC-7901 cells in nude mice. Methods The SGC-7901 cells suspension was injected into the peritoneal cavity of 27 nude mice (2×10^6 cells/mouse ) , then the mice were randomly divided into three groups (9 cases per group ). Control (A) group, the mice was laparotomy only; and CO2 pnemumoperitoneum was established with pressure 5 mmHg (group B ) and 8 mmHg (group C ), the treatment time was 1 h. The control group underwent a midline incision, which was closed after 1 h. Four weeks later, all of the nude mice were killed, and the number of macroscopic abdominal cavity nodules, the total body weight, the mean max diameters and distributing ranges of tumor mass were observed and measured. The rate of viscera ( liver, spleen, peritoneum and mesentery ) metastasis was also recorded in each group, and the Hpa mRNA expression in tumor tissue was measured by RT-PCRo Results The number of abdominal cavity nodules in each group was not significantly different ( control group : 7.4±2.5 ,5 mmHg group: 7.3±2.4, 8 mmHg group: 7.5±2. 1. P 〉0.05). also the total body weight, the mean max diameters and distributing ranges of metastasis showed no difference between each group (P 〉 0.05 ). The Hpa mRNA expression in gastric cancer tissues showed no significant difference in each subgroup 〈 P 〉 0.05 ). Conclusions CO2 pnemumoperitoneum do not accelerate the SGC-7901 cells growth invasion and metastasis in vivo, also it had no significant effect on Hpa mRNA expression.
