白藜芦醇增强顺铂对肝癌细胞Bel-7402的生长抑制作用 被引量:11

摘要 目的观察白藜芦醇与顺铂联合应用对人源肝癌细胞Bel-7402生长的影响。方法采用MTT法、协同指数分析及裸鼠移植瘤模型研究白藜芦醇与顺铂合用对Bel-7402体外增殖及体内生长的抑制作用。结果细胞活性分析表明,两药合用对体外培养的Bel-7402细胞增殖有协同抑制作用,协同指数从0.904-0.739之间分布;动物实验也证实白藜芦醇与顺铂合用可明显抑制Bel-7402裸鼠移植瘤的生长,其药效优于两药单用(P〈0.01)。结论白藜芦醇与顺铂合用可协同抑制人肝癌细胞Bel-7402的生长。 Objective To investigate the effect of resveratrol on the proliferation of human hepatoceiiuiar carcinoma Bel-7402 cells. Methods The effects of combined resveratrol and cisplatin on proliferation of human hepatoceilular carcinoma cell line Bel-7402 were examined in vitro and in vivo. The viability of cultured Bel-7402 cell was tested by MTT assay and the combination index (CI) analysis was performed. Tumor size in xenograft nude mice was also measured. Results Synergistic cytotoxicity was found in Bel-7402 cells measured by MTT assay. The CI values ranged from 0. 904 to 0. 739 for the combination of resveratrol and cisplatin. Studies in nude mice showed that the tumor size in combination therapy was smalle than that with the drug used alone (P〈0.01). Conclusion The combination of resveratrol and cisplatin has synergistic inhibitory effect on the proliferation of hepatocellular carcinoma Bel-7402 cells.
