摘要
目的建立药用植物日本毛连菜中多糖的含量测定方法。方法采用水提醇沉法从日本毛连菜中提取得到毛连菜粗多糖,脱色,用苯酚-硫酸法测定日本毛连菜中多糖含量。结果在3.03-15.10μg/ml范围内具有良好的线性关系,本法平均回收率为(97.55±2.26)%,RSD为2.3%。测定日本毛连菜提取物中总多糖含量以葡萄糖计平均为(5.80±0.06)%。结论该方法简便、准确、可靠,可以用于日本毛连菜中多糖的含量测定。
Objective To establish a method for determining the content of polysaccharide in Picris japoncia Thunb. Methods Poly- saccharide was extracted with water, purified with alcohol precipitation and decolored with H2O2. Then the content of polysaccharide was determined by phenols-sulfuric acid method. Results The linear range for polysaccharide was 3.03 - 15.10 μg/ml. The average re- covery was (97.55 ± 2.26 ) %. RSD was 2.3 %( n = 6 ). The content of polysaccharide in Picris japoncia Thunb was ( 5.80 ± 0.06 ) % (glucose). Conclusion The proposed method is convenient, economic, accurate and reproducible, and can be used to determine the polysaccharide in Pieris japoncia Thunb.
