摘要
木瓜蛋白酶作为外用药,可水解坏死组织,但有副作用。固定化木瓜蛋白酶涂膜剂是由虾壳中提取的壳聚糖与戊二醛交联、固定化制备出来的。0.4%的戊二醛与1%中等粘度壳聚糖(η=100~250MPa·s)经摇床振荡(200r/min,2h)充分交联,加木瓜蛋白酶液(2.09mg/mL,pH7.8)4℃固定12h,涂抹皮肤自然成膜。此固定化涂膜剂最适温度55℃,最适pH7.8,最大载酶量为45mg/g载体,酶相对活力30~35%。与未固定化木瓜蛋白溶液酶相比,提高了其对热、乙醇、脲的稳定性。
Papain is remedy for zymolsising decayed organization,but it has some byeffect. Papain can be immobilized on chitosan and crosslinked with glutaraldehyde: shaking 1% chitosan (η=100~250MPa·s) with 0.4% solution of glutaraldehyde for two hours with a shaketable (200r/min). If 2.09mg/ml papain, pH 7.8 is added and immobilized on the chitosan at 4℃ for 12 hours, papain can be smeared over skin into membrane. The immobiliz ed papain has optimum temperature and optimum pH 55℃ and 7.8 respectively. The maximum load of enzyme is 45mg/g chitosan. Engzyme activity retention is 30~35%. stability of immobilized papain to temperature,alcohol,urea is increased obviously comparred to that of soluble papain. Papain is immobilized on chitosan in order to improve some byeffect of soluble papain.
