摘要
目的观察胰岛素样生长因子-T(IGF-1)基因转染对胚胎干细胞(ES细胞)诱导分化的心肌细胞中IGF-1、肌球蛋白(myoglobulin)、肌动蛋白(actin)及心肌特异性肌钙蛋白-T(Troponin-T)表达的影响。方法将含有IGF-1基因的真核质粒转染入大鼠胚胎干细胞诱导分化的心肌细胞中,采用免疫组织化学方法对心肌细胞内上述4种蛋白的表达进行检测。利用HPIAS-2000图像分析系统测定4种蛋白在各组中表达的平均吸光度值和平均阳性面积率。结果IGF-1基因转染后的心肌细胞内上述4种蛋白的表达明显高于未转染的对照组(P<0.05)。结论IGF-1基因转染的心肌细胞较未转染的心肌细胞合成IGF-1、肌球蛋白、肌动蛋白及心肌特异性肌钙蛋白-T的能力增强,为IGF-1基因治疗心肌梗死提供了实验依据。
Objective To observe the effects of IGF-1 gene transfection on the expression of IGF-1,myoglobulinactin,actin,and Troponin-T in myocardial cells differentiated from embryonic stem cells. Methods Myocardial cells differentiated from embryonic stem cells of rats were transfected with eukaryotic plasmid containing IGF-1 gene. The expression levels of IGF-1, myoglobulinactin, actin, and Troponin-T proteins in myocardial cells were detected by using immunohistochemistry on the 1 st, 2nd, 3rd, 5th and 7th day after transfection. HPIAS-2000 image analysis system was applied to measure the average absorbance(A) values and the average positive area rate of IGF-1, rnyoglobulinactin, actin, and Troponin-T proteins in each group. Results The expression levels of IGF-1, myoglobulin, actin and Troponin-T proteins were significantly higher in IGF-1 gene transfected group than those in control group(P〈0.05). Conclusion The synthesis of IGF-1,myoglobulin,actin and Troponin-T in myocardial cells transfected with IGF-1 gene was promoted,which provided a theoretical foundation for the IGF-1 gene therapy of myocardial infarction.
