ERK在大鼠浅筋膜中的表达及针刺后的改变特征 被引量:4

摘要 目的探讨针刺对SD大鼠浅筋膜层的细胞外调节蛋白激酶(ERK1/2)表达情况的影响。方法18只SD大鼠随机分为6组,对照组和针刺1、2、3、4、5组,每组大鼠3只。针刺各组在大鼠腹股沟区给予电针治疗后,分别在针刺后0、1、6、12和36h等5个时间点取以针刺点为圆心,直径约1.5cm处的浅筋膜,对照组也在针刺点相应的部位和足三里穴区取材。用Western blot蛋白免疫印迹方法检测各组的ERK1/2及磷酸化ERK1/2(p-ERK1/2)的表达情况。结果穴位和非穴区的浅筋膜的ERK1/2和p-ERK均有表达,但两者未见明显差异,针刺后各组针刺侧的ERK1/2的表达均明显高于自身对照侧。结论正常的疏松结缔组织中可能通过ERK细胞信号转导蛋白的磷酸化来参与组织的增殖和分化;而针刺对于ERK信号转导有促进作用,这也可能是针灸对机体的病理和生理状态进行调控的机制之一。 Objective To observe the effect of acupuncture on the expression ofextracellular signal-regulated protein kinases 1/2 (ERK1/2) in the subcutaneous fascia of SD rats. Methods Eighteen SD rats were randomly divided into 6 groups (n=3) including 5 acupuncture groups and a control group. The rats in the 5 acupuncture groups received electro-acupuncture therapy in the regions of the inguinal groove, and at 0, 1, 6, 12, and 36 h after the last therapy, the superfacial fascia surrounding the acupuncture point (about 1.5 cm in diameter) were collected. The fascia tissues at the corresponding sites and at the acupoint Zusanli (ST36) were obtained from the control rats. The expression of ERK1/2 and phosphorylated ERK1/2 (p-ERK1/2) in the tissues were detected by Western blotting. Results ERK1/2 and p-ERK1/2 expressions were detected in the tissues harvested from both the acupoint and the non-acupoint in the control rats with similar expression intensities. In the rats of each acupuncture group, ERK1/2 expression was significantly increased on the acupuncture side in comparison with the control side. Conclusion The normal loose connective tissue may participate in tissue proliferation and differentiation possibly via phosphorylation of ERK. Acupuncture can promote the signal transduction pathway of ERK, which can be a possible mechanism for the effect of acupuncture in modulating the physiopathological conditions.
