摘要 以濒危果树野生欧洲李当年生新梢为外植体,进行组织培养研究,以期筛选出最优培养基配方及培养程序。结果表明:野生欧洲李初代培养时,以B5为基本培养基优于MS培养基;培养基中添加一定量激素的外植体生长势明显优于不添加激素;以B5为基本培养基,6-BA质量浓度0.5~1.0mg/L,并添加一定量的生长素(IBA、NAA)效果较优,能较快启动生长反应,并初步形成一定的增殖率;芽增殖培养阶段,6-BA质量浓度为0.5~1.0mg/L时,以NAA为生长素,NAA和6-BA适宜的比例为1:(1~3),以IBA为生长素,IBA和6-BA适宜的比例为1;(3~5)较适宜。 In order to select out optimum media formula and culture program, tissue culture techniques of wild European plum (Prunus dornestica L. ) were researched, which belong to endangered fruit tree. The results showed that Bsbasic medium was better than MS medium in initial culture; Explants had better growth vigor in the media supplemented with some hormones than that in the media without any hormone ; Explants started to grow more quickly in Bs supplemented with 6-BA 0. 5-1.0 mg/L and growth hormone (NAA or IBA), and some proliferation buds could be gained; The suitable concentration ratio of NAA to 6-BA was 1 : (1--3) in the media supplemented with 6-BA 0.5--1.0 mg/L and NAA for proliferation of buds, and that of IBA to 6-BA was (1 : 3--5) in the media supplemented with 6-BA 0. 5-1.0 mg/L and IBA.
