山羊MyoG基因Csp6I酶切多态性分析 被引量:10

摘要 采用限制性内切核酸酶Csp6I对山羊MyoG基因的959 bp扩增产物进行PCR-RFLP多态性分析,检测到3种基因型,其酶切位点分别由2种共显性基因控制;对该片段的纯合型(AA、BB)分别测序发现,BB型在1791位点发生了T→C的突变。上述结果为首次试验证实山羊MyoG内含子二区域存在多态性。 MyoG gene in goat was amplified and an uniform fragment of 959bp was obtained. The 959bp PCR product was digested with restriction endomuelease Csp6Ⅰ and genetic polymorphism was investigated by PCR - RFLP. Three kinds of genotypes were found in these populations, which were controlled by two alleles. The amplified fragments of AA and BB of the fragment were sequenced. The result showed that 959 bp BB genotype had one mutation at 1791 site(T→C). The result above -mentioned has confirmed for the first time that there were polymorphisms in intron 2 region of MyoG gene.
