摘要 综述糖原合成酶激酶-3β与2型糖尿病、阿尔茨海默病、肿瘤等疾病发生发展的关系及其抑制剂的研究和开发。糖原合成酶激酶是普遍存在于真核细胞中的一种多功能丝氨酸/苏氨酸激酶,参与多条细胞信号传导通路,其作用主要包括调节糖原的合成代谢、细胞的分化与增殖以及基因表达,它的活性异常与多种疾病的发生发展有着密切的关系,已成为备受关注的药物靶点。 The correlationship of glycogen synthase kinase-3β(GSK-3β) with these diseases, type 2 diabetes mellitus, Alzheimer's disease, tumor and so on, and the research and development of GSK-3β inhibitors were reviewed. GSK-3β commonly found in eukaryote is a multifunctional serine/threonine protein kinase, which is involved in a number of cell signalling pathways, playing the important roles in glycogen metabolism, cell differentiation and proliferation and gene expression. There is a close correlation of the abnormal activity of GSK-3β with many diseases and thereby it has become a well-interested drug target.
