摘要
目的观察妇科活血止痛颗粒对急性血淤模型大鼠血液流变学的影响。方法雌性SD大鼠50只,随机分为空白对照组,急性血淤模型组,妇科活血止痛颗粒低剂量组(0.23g/kg)、中剂量组(0.46g/kg)和高剂量组(0.92g/kg),每组10只,全部动物以标准饲料喂养,3次/d,对照组和血淤模型组大鼠每日末次喂食后1h以10mL/kg生理盐水灌胃,低、中高剂量组每日末次喂食1h后按0.23、0.46、0.92g/kg剂量给予妇科活血止痛颗粒混悬液灌胃,灌胃容量10mL/kg,共10 d。第10天给药1 h后,除对照组外的其他组动物均皮下注射肾上腺素,冰水浸泡建造急性血淤模型,造模结束18h后,股动脉取血测定不同切变率的全血黏度、血浆黏度、红细胞压积、红细胞聚集指数等指标。结果血淤模型组大鼠全血黏度、全血还原黏度(低切和中切)明显高于对照组(P<0.05或P<0.01);低中剂量组明显低于血淤模型组(P<0.05或P<0.01);血淤模型组大鼠红细胞压积、红细胞聚集指数明显高于对照组(P<0.05或P<0.01),红细胞压积低、中、高剂量组大鼠均低于血淤模型组(P<0.05或P<0.01),红细胞聚集指数积低、中剂量组大鼠均低于血淤模型组(P<0.05或P<0.01)。结论妇科活血止痛颗粒对改善大鼠血液流变学状态具有良好作用。
Objective To observation the effect of Fukehuoxuezhitong granule on hemorheology in acute blood stasis model rats. Methods 50 female SD rats were randomly divided into five groups: normal control group, acute blood stasis model group, Fukehuoxuezhitong granule of low-dose group (0.23g/kg), middle-dose group (0.46g/kg) and high-dose group (0.92g/kg), all animals fed a standard diet, 3 times/d, the control group and blood stasis model rats fed to 10ml/kg intragastric administrated saline 1 h after the last day. low-, middle- and high-dose group fed 0.23,0.46,0.91g/kg doses of Fukehuoxuezhitong granule lh after, by gavage capacity 10mL/kg, a total of 10 d. 1 h after the last day, with the exception of the control group , other group of animals were made by soaked ice water after injected adrenaline hydrochloride in a major dose. The blood viscosity (high, middle, low shears), blood re- duce viscosity, plasma viscosity, hematocrit and erythrocyte aggregation index were measured with Auto-Viscometer. Results Compared with control group, blood stasis model group showed middle,low shear viscosity increased sig- nificantly ( P 〈 0.05 or P 〈 0.01 ), middle- and low-dose groups had a decline in the enhancement of whole blood viscosity in rats with blood stasis (P 〈0.05 or P 〈0.01). the hematocrit and the erythrocyte aggregation index of the blood stasis model group Significantly increased as compared with control group( P 〈 0. 05 or P 〈 0. 01), In the low-, middle- and high-dose groups, the hematocrit and the erythrocyte aggregation index decreased ( P 〈 0. 05 or P 〈 0.01). Conclusion Fukehuoxuezhitong granule has the fine effect to improving rat on hemorheology.
