电压门控性钠离子通道β_1亚基条件性过表达转基因小鼠的制备

摘要 目的制备电压门控性钠离子通道β1亚基基因(SCN1b)条件性过表达转基因小鼠。方法将电压门控性钠离子通道β1亚基的编码序列片段插入表达载体pCMV-loxp-lacZ-stop-loxp-IRES-eGFP中;应用显微注射法将构建好的目的载体注入供卵小鼠受精卵核中,移植受精卵入假孕雌鼠输卵管;经PCR检测阳性的后代小鼠即为原代小鼠,通过后代小鼠X-gal染色鉴定目的基因在原代小鼠的整合部位。结果共获得13只仔鼠,其中3只为目的基因表达阳性的原代小鼠,其中1只原代小鼠的背根神经节(DRG)细胞X-gal染色呈阳性。结论成功制备在DRG条件性过表达SCN1b的转基因小鼠模型,为深入研究电压门控性钠离子通道β1亚基在痛觉调控中的功能提供了工具鼠。
