HPK1在缺糖缺氧诱导的大鼠海马神经元凋亡中的作用

摘要 目的观察造血祖细胞激酶1(HPK1)对缺糖缺氧(OGD)诱导的大鼠海马神经元凋亡的作用。方法采用细胞免疫荧光技术通过免疫荧光显微镜和激光共聚焦显微镜观察HPK1是否在大鼠脑细胞中存在;采用DAPI染色技术观察HPK1反义寡核苷酸对OGD诱导的海马神经元凋亡的作用。结果在体外培养的原代大鼠海马神经元中有HPK1的表达。HPK1反义寡核苷酸对OGD诱导的海马神经元凋亡具有保护作用,并有剂量依赖性。结论HPK1反义寡核苷酸对OGD诱导的海马神经元凋亡具有保护作用。 Objective To investigate the role of HPK1 in hippocampus neuron apoptosis induced by oxygen - glucose deprivation (OGD). Methods Immunofluorescence technique was used to investigate whether there was HPK1 in hippocampus neuron under the fluorescence microscope and the confocal microscopy. DAPI staining was used to investigate the role of HPK1 antisense oligodeoxynucleotides in hippocampus neuron death induced by OGD. Results It was found that HPK1 were expressed in rat hippocampus neurons cultured in vitro and administration of HPK1 antisense oligodeoxy- nucleotides could significantly decrease the apoptosis of hippocampus neurons which induced by OGD. Conclusion Knock - down of HPK1 ; expression provides neuroprotection against the apoptosis of hippocampus neurons which induced by OGD.
