摘要
目的通过比较肾上腺素和重组人粒细胞集落刺激因子(rhG-CSF)单独或联合应用动员小鼠外周血造血干祖细胞(HSPC)的效果,以了解肾上腺素对HSPC的动员作用,为临床制定更为优良的动员方案奠定实验方面的基础。方法BALB/c小鼠48只,随机分成4组,每组12只。A组作为对照组,皮下注射生理盐水(NS)100μl.d-1;B组皮下注射rhG-CSF 250μg.kg-1.d-1;C组腹腔注射肾上腺素2.5 mg.kg-1.d-1;D组皮下注射rhG-CSF 250μg.kg-1.d-1+腹腔注射肾上腺素2.5 mg.kg-1.d-1(rhG-CSF后3 h),各组均连续用药5天。动态观察各组小鼠外周血中白细胞数(WBC)、Lin-c-kit+(KL)、Lin-c-kit+Sca-1+(KSL)细胞比例的变化。结果D组外周血WBC数及KL、KSL细胞比例明显高于B组(P<0.05)或C组及A组(P<0.01);B组外周血WBC数及KL、KSL细胞比例明显高于C组及A组(P<0.01);C组外周血WBC数明显高于A组(P<0.01),但KL、KSL细胞比例与A组比较无显著性差异(P>0.05)。结论肾上腺素可协同rhG-CSF参与HSPC的动员。
Objective To investigate the mobilizing effect of epinephrine on HSPC by comparative study on the effects of epinephrine and recombinant human granulocyte colony stimulating factor ( rhG - CSF) when used alone or in sequential combination in mobilizing hematopoietic stem and progenitor cells (HSPCs) in mice and optimize the strategy of mobilization. Methods 48 BALB/c mice were randomly divided into 4 groups, with 12 mice in each group. In Group A (the control group), each mice was given a subcutaneous injection of NS 100μl · d^-1; in Group B, each mice was subcutaneously injected with rhG - CSF 250 μg · kg^-1·d^ -1 alone ; in Group C, each mice was intraperitoneaily injected with epinephrine 2.5 mg ·kg^-1·d ^-1 alone ; in Group D, each mice was injected in sequential combination of rhG - CSF 250 μg·kg^-1·d^-1 subcutaneously and epinephrine 2.5 mg·kg^-1·d^-1 intraperitoneally 3 h after rhG - CSF. All the groups were injected for five consecutive days. Variations of the white blood cell (WBC) count, and the proportions of KL and KSL cells in peripheral blood of mice were dynamically observed. Results The WBC count, and the proportions of KL and KSL cells in peripheral blood of Group D were all markedly higher than those of Group B ( P 〈 0. 05 ) and Group C or Group A ( P 〈 0.01 ) ; Group B mobilized more WBC, KL and KSL ceils in peripheral blood as compared to those of Group C and Group A (P 〈 0. 01 ) ; Group C had much more yield of WBC than that of Group A ( P 〈 0.01 ) , while the proportions of KL and KSL cells had no significant difference (P 〉 0.05). Conclusion Epinephrine can participate in mobilizing HSPC in coordination with rhG - CSF.
