木犀草素对T细胞IL-2mRNA、INFγmRNA表达的影响 被引量:6

摘要 目的:研究木犀草素(luteolin)对佛波醇酯(Phorbol ester,PMA)+离子霉素(Ion)体外诱导的小鼠T细胞活化和增殖的影响。方法:以佛波醇酯+离子霉素刺激小鼠脾淋巴细胞来源的T细胞,以不同终浓度的木犀草素与T细胞共培养,用MTT法测定细胞增殖;采用反转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)法检测药物对PMA刺激作用下的T细胞中Th1/Th2细胞因子IL-2mRNA与INFγmRNA表达的影响。结果:PMA+Ion刺激48h后,小鼠T淋巴细胞增殖显著高于对照组(P<0.01);木犀草素(10、20、30μM)可显著降低PMA引起的细胞增殖(P<0.01)。PMA+Ion刺激T细胞6h后,细胞IL-2mRNA、INFγmRNA表达与空白组比较明显增强(P<0.01),木犀草素(10、20、30μM)能抑制PMA+Ion刺激的T细胞IL-2mRNA、INFγmRNA的表达(P<0.01);结论:木犀草素抑制小鼠T细胞的增殖和活化,可能与抑制IL-2mRNA、INFγmRNA的表达有关,提示木犀草素具有机体免疫调节作用。 Objective: To investigate the effect of luteolin on the activation and proliferation of mouse T cells stimulated by the combination of PMA plus Ion in vitro. Methods: T cells from mouse were stimulated by PMA plus Ion and incubated with different concentrations of luteolin, followed by the cell proliferation assay by MTT and detection of IL-2mRNA and INFγmRNA in T cell by RT-PCR. Results: After the stimulation by PMA plus Ion for 6h, T cell proliferated markedly when compared with control group (p〈0. 01); luteolin (10, 20, 30μM) degraded the proliferation oft cells significantly when induced by PMA plus Ion (p〈0.01); the cytoline expression of IL-2mRNA, INFγmRNA of T cells were inhibited notably by various concentrations (10, 20, 30μM) of luteolin when compared with control group (p〈0.01). Conclusions: Luteolin can prevent the activation and proliferation of mouse T cells, which is probably achieved by inhibiting the expression of IL-2mRNA and INFγmRNA.
