摘要
目的观察葡萄糖耐量因子(GTF)葛根功能牛奶对葡萄糖耐量减低(IGT)小鼠的干预作用,以及对骨骼肌葡萄糖转运蛋白-4(GLUT-4)基因表达的影响。方法采用D-半乳糖腹腔注射法诱导的IGT小鼠灌胃,观察空腹血糖(FBG)、葡萄糖耐量试验(OGTT)以及后2h血糖(2hBG)含量的效应,并通过RT-PCR半定量法测定骨骼肌GLUT-4基因表达。结果GTF葛根功能牛奶能抑制2hBG升高,增加GLUT-4基因表达,与模型对照组比较有显著性差异(P<0.01)。结论GTF生物活性牛奶添加葛根素后能明显增强降糖作用,产生了类似中医方剂学相须配伍的效应,其机制有待进一步研究。
Objective To observe the intervention of GTF puerarin functional milk on impaired glucose tolerance(IGT)mice and the impact of it on glucose transport proteins-4(GLUT-4)gene expression in skeletal muscle.Methods Stomach perfusion was applied to IGT mice induced by D-galactose peritoneum injection.It was to observe the impacts on fasting blood glucose(FBG)and oral glucose tolerance test(OGTT)as well as 2-hour blood glucose(2 h BG).RT-PCR semi-quantitative method was used to examine GLUT -4 gene expression in skeletal muscle. Results GTF puerarin functional milk suppressed the increase of 2 h BG and improved GLUT -4 gene expression, which indicated significant difference compared with those in model group ( P 〈 0.01 ). Conclusion GTF bioactive milk enhances apparently the action on reducing glucose after puerarin supplement, which produces similar effects as mutual promotion of formula compatibility in Chinese medicine. But a further study is anticipated on its mechanism.
