摘要
抗氧化蛋白酶在洗涤剂工业有重要应用价值。天然环境中可以筛选到产抗氧化蛋白酶的菌株,但分析抗氧化酶的基因无法找到抗氧化规律。定点突变法可以增强普通洗涤蛋白酶的抗氧化性,但活性降低。抗氧化蛋白酶空间结构的研究结果,尚未能阐明抗氧化机理及氧化后底物专一性改变的原因。研究抗氧化蛋白酶对酶制剂的研发有重要指导意义。
Oxidatively stable proteases (OSP) are of important value as detergent additives. Many OSP-producing strains are screened from the environment, but the number of the OSP genes is still scanty and the alignment of the present eight OSPs does not demonstrate the common characters amongst the amino acid sequences. Oxidation-resistant detergent protease mutants can be obtained at the cost of the activities by using site-directed mutagenesis technique. Although it is inferred that the slow oxidation of Met in the vicinity of the catalytic Set is a possible mechanism underlying the oxidative stability of KP-43, the detailed mechanism is still puzzling. Perspective studies are required to investigate the theoretical problems on OSP which will bring further insights of the relationship between the protease structure and function. The results will provide clues to research and develop detergent products of OSP.
