摘要
研究水稻品种蛋白质亚基的百分含量,探讨水稻谷蛋白(PB-Ⅱ)亚基和醇溶蛋白(PB-I)亚基的相关性。以日本现行的SDS-PAGE凝胶电泳检测方法,利用Labwork4.5电泳凝胶成像数据处理分析系统,对水稻不同品种进行全蛋白电泳及蛋白质亚基检测及进行电泳凝胶谱带13kDa醇溶蛋白单一亚基含量的分析。试验结果表明,龙粳8号的醇溶蛋白亚基含量较低为11.53%;富士光和松99-135的谷蛋白亚基含量较高为72.87%和69.52%;绥粳3号的醇溶蛋白亚基含量相对较高为15.25%。醇溶蛋白与PB-Ⅱ谷蛋白显著负相关,相关系数R=-0.977315。16kDa醇溶蛋白亚基与PB-Ⅱ谷蛋白极显著负相关,相关系数R=-0.994356。13kDa醇溶蛋白亚基与PB-Ⅱ谷蛋白显著负相关,相关系数R=0.913779。绥粳3号和籼粳交的特优21在单一亚基分析中,其13kDa醇溶蛋白亚基含量明显高于其他几个品种。也就是说,不同水稻品种的蛋白质亚基含量不同;不同品种的13kDa醇溶蛋白亚基含量存在差异;醇溶蛋白总量、13kDa、16kDa醇溶蛋白亚基与PB-Ⅱ谷蛋白负相关。
The present study is to discuss the relationship of glutelin (PB-II) subunits and prolamin (PB-I) subunits through investigating subunits content of protein in different rice varieties. The whole protein electrophoresis, detection of protein subunits and gel electrophoresis bands of the single 13 kDa prolamin subunit content in different varieties of rice were analyzed by SDS-PAGE. The results showed that the content of prolamin subunit in Longjing 8 was lower (11.53%), whereas the content of prolamin subunit in the 3rd Sui Japonica was higher (15.25%). Rice varieties, such as 99-135 and the Fujitsu-Zein, had relatively higher glutelin subunit content. The content was 72.87% and 69.52%, respectively. The content of prolamin and 13 kDa prolamin subunit had significant negative correlation with glutelin content. The correlation coefficient was-0.977315 and-0.913779, respectively. The relationship between 16 kDa prolamine subunit content and prolamin content was very significant negative .The coefficient was-0.994356. According to a singlel3kDa subunit analysis of the rice's varieties, the 13 kDa prolamin subunit content was obviously higher than that of several other varieties in the 3rd Sui Japonica and Indica and Japonica intercourse Teyou 21. In other words, the different varieties of rice have the different protein subunits content. The content of 13 kDa prolamin subunit is different in different varieties. Prolamin content, 13kDa prolamin subunit content and 16kDa prolamin subunit content have significant negative correlation with glutelin content.
