Detection of MAGE_(A3) mRNA and hSTC_1 mRNA in Peripheral Blood of Patients with Early Stage Cervical Cancer and Its Clinical Significance

摘要 目的研究早期宫颈癌患者外周血中MAGE。和hSTC1mRNA的表达及临床意义。方法采用RT-PCR法检测60例ⅠA~ⅡA期宫颈癌患者及15例同期入院的排除其他恶性肿瘤的子宫肌瘤患者外周血中MAGEA3和hSTC,mRNA的表达,并分析其与临床病理因素之间的关系。结果60例早期宫颈癌患者外周血中MAGEA3和hSTC1mRNA的阳性表达率分别为46.67%(28/60)和40%(24/60),子宫肿瘤均为0%。MAGEA3和hSTC1mRNA的单独阳性表达率和双阳性表达率除与淋巴结转移和脉管瘤栓显著相关外(P〈O.05),与宫颈癌的预后因素如年龄、分期、分化、组织类型、肿瘤大小均无关(P〉0.05),但随着肿瘤分期及肌层浸润深度的增加有逐步上升的趋势。结论MAGEA3和hSTC1可作为检测早期宫颈癌外周血微转移的生物学指标,但对预后的意义有待于进一步研究。 Objective To study the expression of MAGEA3 and hSTC1 mRNA in peripheral blood of patients with FIGO Stage ⅠA to ⅡA cervical cancer and to investigate its clinical significance. Methods Using the reverse transcription polymerase chain reaction (RT-PCR),MAGEA3 and hSTC1 mRNA expression was examined in peripheral blood from 60 patients with early cervical cancer before operation. As control, 15 patients with uterine myoma were also investigated. Results The positive rate for MAGEA3 and hSTC1 mRNA expression were 46.7%(28/60) and 40%(24/60) in 40 cervical cancer respectively,0% and 0% in uterine myoma. The expression of MAGEA3 or hSTCI mRNA were significantly associated with lymph node metastasis and lymphvascular involvement(P 〈 0.05), but not with prognostic factors including stage,differentiation,pathological types,bully turnout size,stromal invition,same to the expression of both two. Conclusion MAGEA3 and hSTC1 may be potential biomarkers for detecting micrometastasis in peripheral blood of patients with early stage cervical cancer,but the predictive value needs further studies.
