摘要
目的研究三甲益肝颗粒(SJG)对四氯化碳(CCl4)诱导的实验性大鼠肝纤维化的抑制作用,并探讨其可能的作用机制。方法健康SD大鼠50只随机分为3组。模型组(18只)和SJG组(17只)均采用40%CCl4花生油溶液皮下注射诱导大鼠肝纤维化模型,每周2次;对照组(15只)皮下注射生理盐水。从实验开始之日起,模型组给予蒸馏水灌胃,SJG组每天给予SJG灌胃。分别于6周末、8周末收获大鼠肝组织做Masson染色,检测胶原纤维的表达;免疫组化检测α-平滑肌肌动蛋白(α-SMA)的表达。结果模型组胶原纤维所占面积多于对照组(P<0.01),SJG组胶原纤维所占面积少于模型组(P<0.01)。正常对照组大鼠肝组织α-SMA仅在汇管区的小叶间动脉、静脉血管壁处表达,模型组α-SMA的表达多于正常对照组(P<0.01),SJG组α-SMA的表达较模型组减少(P<0.01),8周α-SMA的表达虽比6周有所增加,但差异无统计学意义(P>0.05)。结论SJG可能抑制肝星形细胞的活化,抑制胶原纤维的沉积,从而有效地减轻肝纤维化的程度。
Objective To investigate the inhibitive effect of Sanjiayigan granule (S-JG) on carbon tetrachloride (CCl4) - induced hepatic fibrosis in rats and analyse the possible mechanism in preventing experimental hepatic fibrosis by S-JG. Methods 50 healthy male rats were divided into three groups. Model group (n = 18) and SJG group (n = 17) were treated with 40% CCl4 solution diluted by peanut oil through subcutaneous injection two times a week to establish hepatic fibrosis model. The rats of control group were only treated with subcutaneous injection of normal saline (n = 15). Since the first day of the test, the rats were given medicine or distilled water respectively. Hepatic tissue of rats were collected at the end of 6th and 8th week respectively. The collagen fibers were examined by Masson stain. The expressions of α- smooth muscle actin (α- SMA) were detected by immunohistochemical method. Results (1)The area of collagen fibers in the model group increased significantly compared with control group(P〈0.01). The area of collagen fibers in the S- JG group decreased significantly compared with the model group(P〈0.01). (2)The α - SMA only expressed in the vessel wall of interlobular artery and vein in control group. The expression of α - SMA in the model group increased significantly compared with control group(P〈0. 01). The expression of α - SMA in S- JG group decreased significantly compared with the model group(P〈0.01). At the end of 8th of week the expression of α- SMA increased compared with that at the end of 6th of week,but the difference was not significant(P〉0.05). Conclusion The S- JG can inhibit the expression of α - SMA on liver of hepatic fibrosis rats. It may inhibit the activation and proliferation of hepatic stellate cells and decrease the degree of experimental hepatic fibrosis of rats efficaciously.
