摘要
目的观察不同剂量、给药时间点及给药方式的异丙酚对全脑缺血大鼠海马CA1区锥体细胞的保护作用。方法80只雄性Wistar大鼠按照不同的给药剂量、给药时间点及方式随机分组。采用4血管闭塞法制造大鼠全脑缺血模型,硫堇染色下对锥体细胞的死亡进行评估。结果异丙酚可剂量依赖性地保护锥体细胞,给药时间点越接近脑缺血,异丙酚的神经保护作用越明显。持续静脉输注异丙酚的神经保护作用较好。结论在脑缺血前后的"有效时间窗"内,静脉持续输注麻醉剂量的异丙酚可有效地保护神经元。
Objective The study was undertaken to investigate the neuronal protective effect of propofol administrated with different doses, time points and protocols against delayed neuronal death (DND) of pyramidal neurons in the CA1 hippocampus induced by global brain ischemia.Methods Eighty male Wistar rats were randomly divided in to several groups according to different doses, time points and protocols of the administration.Global brain ischemia was produced by the four vessels occlusion method.DND of pyramidal neurons in the CA1 hippocampus was examined with the neuropathological evaluation under thionin staining.Results No lower than 50mg/kg -1 of propofol could protect pyramidal neurons against DND of pyramidal neurons in the CA1 hippocampus.Propofol might produce better protective effect on the neurons when used within a short period not out the "treatment window" before or after ischemia insult,and with a protocol consisted of an initial bolus intravenous injection and following persistent intravenous infusion.Conclusion The results suggested that doses, time points and protocols of propofol administration could result in influences on the neuronal protection of propofol.
