摘要
目的探讨内源性甲状旁腺激素在实验性小鼠牙周炎牙槽骨吸收过程中的作用。方法大肠杆菌内毒素脂多糖牙龈局部注射法建立小鼠实验性牙周炎牙槽骨吸收模型,通过组织形态学、骨总胶原染色、耐酒石酸酸性磷酸酶染色等方法,比较分析甲状旁腺激素基因敲除小鼠和野生型小鼠牙槽骨吸收的差异。结果和野生型小鼠相比,甲状旁腺激素基因敲除小鼠的牙周骨丧失值、牙槽骨内单位长度骨小梁破骨细胞数以及骨面上破骨细胞的总长度占骨面长度的百分比则明显增加,牙周单位面积骨量明显降低。结论内源性甲状旁腺激素在实验性牙周炎牙槽骨吸收过程中起着重要的抑制作用。
Objective To study the effects of parathyroid hormone on periodontitis-associated bone absorption in mice. Methods In this protocol, adult mice were used to induce periodontitis-associated bone absorption by injection of the bacterial endotoxin lipopolysaccharide(LPS) into the maxillary gingiva of the first molar. The differences between the wild-type and parathyroid hormone gene knockout mice were assessed by histomorphology, total collogen staining and tartrate-resistant acid phospbatase staining. Results The PBL, the N. Oc/B. Pm and Oc. S/B. S were increased significantly, but the BV/TV of the alveolar bone were reduced in parathyroid hormone gene knockout mice, compared with those in the wild-type littermates. Conclusion These studies suggest that endogenous parathyroid hormone is essential in the inhibition of pefiodon- titis-associated bone absorption in mice.
