摘要
目的研究芎汤不同极性部位抗小鼠急性心肌缺血的药效作用。方法制备芎汤超临界CO2萃取部位、石油醚部位、乙酸乙酯部位及正丁醇部位(简称SFE部位、Pet部位、EtOAc部位、BuOH部位),通过高效液相色谱指纹图谱对其所含成分进行表征;用腹腔注射异丙肾上腺素造成小鼠急性心肌缺血模型,观察不同极性部位对心电图、红细胞膜流动性的影响。结果对急性心肌损伤小鼠,芎汤超临界提取部位、正丁醇部位可拮抗S-T段异常变化;正丁醇部位可降低荧光探剂DPH标记的红细胞膜荧光偏振度,增强红细胞膜流动性。结论芎汤超临界提取部位、正丁醇部位具有一定的抗心肌缺血药效活性。
OBJECTIVE To explore the protective effect of different polar extracts from Xiongqiong Decoction on acute experimental ischemic myocardium of mice. METHODS The carbon dioxide supercritical extract,petroleum ether extract, acetoaeetate extract and n-butyl alcohol extract (part SFE, part Pet, part EtOAc and part BuOH) for Xiongqiong Decoction were prepared in turn. Using high performance liquid chromatography (HPLC), the fingerprint chromatograms for different extracts have been established respectively. Acute myocardial ischemic model induced by intraperitoneal administration of isoproteronol (ISO) in mice was adopted to observe the effects of the different extracts on the electrocardiogram and the membrane fluidity of akaryoeytes. RESULTS Compared with the model group, groups treated with part SFE and part n-BuOH have shown an action of reducing the elevation of ST segment induced by ISO. Part SFE could decrease the fluorescence polarization which demonstrated the effect of increasing the cell membrane fluidity in the inactive state. CONCLUSION Part SFE and part n-BuOH have protective effect on ISO induced acute myocardial ischemia in mice.
