黄精中小分子糖对小鼠免疫功能的影响 被引量:18

摘要 背景:滋补中药黄精的主要有效成分是糖类化合物,炮制加工使黄精中多糖含量降低,而小分子糖类含量增加。为分析小分子糖与黄精补益作用的相关性,课题组对此开展实验。目的:观察黄精中小分子糖对正常小鼠及对环磷酰胺所致免疫功能低下小鼠免疫功能的影响。设计、时间及地点:完全随机分组设计、对照实验,于2008-02/05在河南中医学院动物实验中心完成。材料:选取昆明种小鼠180只,100只用于正常小鼠实验,80只用于环磷酰胺致免疫功能低下小鼠实验。方法:①正常小鼠腹腔巨噬细胞吞噬实验及溶血素、溶血空斑实验:各选用50只小鼠,按随机数字表法分为5组,每组10只,各用药组分别灌服0.9,0.6,0.3g/kg的黄精小分子糖水溶液及阳性对照0.1g/kg的香菇多糖混悬液,空白对照组灌服同体积的生理盐水。②环磷酰胺致免疫低下小鼠腹腔巨噬细胞吞噬实验及溶血素、溶血空斑实验:各选用40只小鼠,按随机数字表法分为4组,即模型组、空白对照组、香菇多糖片0.1g/kg组、0.9g/kg黄精小分子糖组,每组10只。除空白对照组外,其余各组均建立环磷酰胺致小鼠免疫抑制模型。主要观察指标:观察各组小鼠腹腔巨噬细胞吞噬率、吞噬指数及溶血素、溶血空斑的形成情况。结果:0.9,0.6g/kg小分子糖均能显著提高正常小鼠腹腔巨噬细胞对鸡红细胞的吞噬百分率及腹腔巨噬细胞的吞噬指数(P<0.01),促进小鼠溶血素和溶血空斑形成(P<0.01),以0.9g/kg黄精小分子糖组作用为最优。0.9g/kg小分子糖和香菇多糖片可显著提高免疫低下模型组小鼠腹腔巨噬细胞对鸡红细胞的吞噬指数和吞噬百分率(P<0.01),且以0.9g/kg小分子糖组效果明显;0.9g/kg小分子糖组可显著促进模型组小鼠溶血素和溶血空斑的形成(P<0.01)。结论:黄精小分子糖对机体免疫功能具有一定的增强作用,以剂量0.9g/kg作用为最优。 BACKGROUND: The active component of rhizoma polygonati is saccharide compound. Processing reduces the polysaccharide content in the rhizoma polygonati, but increases low molecular weight saccharide. The correlation between low molecular weight saccharide and tonification of rhizoma polygonati is explored. OBJECTIVE: To observe the effect of low molecular weight saccharide in rhizoma polygonati on the immune function of normal mice and immunosuppression ones caused by cyclophosphamide. DESIGN, TIME AND SETTING: Randomized, controlled experiment. The study was performed at the Animal Experimental Center of Henan University of Traditional Chinese Medicine from February to May 2008. MATERIALS: Of 180 Kuming mice, 100 were selected for normal experiment, and the remaining as immunosuppression experiment. METHODS: Peritoneal macrophage experiment, hemolysin and hemolytic antibody plaques formation tests in normal mice: 50 mice selected for each experiment were randomly divided into 5 groups (n=10): blank control (normal saline), positive control (0.1 g/mL Xianggu Duotang) and treatment groups (0.9, 0.6, 0.3 g/kg low molecular weight saccharide of rhizoma polygonatl). Peritoneal macrophage experiment and hemolysin hemolytic antibody plaques formation tests in mice with immunosupression induced by cyclophosphamide: 40 mice selected for each experiment were randomly divided into 4 groups (n=-10): model, blank control, Xianggu Duotang (0.1 g/kg) and 0.9 g/kg low molecular weight sugars in rhizoma polygonati groups. Except for the blank control group, mice in other groups were made into cyclophosphamide-induced immunosuppressional models. MAIN OUTCOME MEASURES: Peritoneal macrophages phagocytic index and percentage, and the hemolysin and hemolytic plaques formation were observed. RESULTS: 0.9 and 0.6 g/kg low molecular weight saccharide in rhizoma polygonati significantly increased the peritoneal macrophages phagocytic index and percentage (P 〈 0.01), significantly promoted the hemolysin and hemolytic plaque formation in normal mice, in particular the 0.9 g/kg group. 0.9 g/kg low molecular weight saccharide and Xianggu Duotang significantly increased the peritoneal macrophages phagocytic index and percentage in cyclophosphamide-induced immunosuppressional mice (P 〈 0.01 ), in particular the 0.9 g/kg group. In addition, 0.9 g/kg low molecular weight saccharide remarkably promoted the hemolysin and hemolytic plaque formation (P 〈 0.01). CONCLUSION: Low molecular weight saccharide in rhizoma polygonati can significantly increase the immune function in mice, in particular at dosage of 0.9 g/kg.
