摘要
目的探讨中药熏洗加电针治疗膝骨性关节炎的机制。方法32只日本大耳兔随机分成4组:空白组、模型组、扶他林组和中药熏洗加电针组,采用手术方法制作兔膝骨性关节性模型,造模成功后中药熏洗加电针组给予中药熏洗加电针治疗4周,扶他林组给予扶他林外搽4周。用免疫组化法检测各组软骨组织基质金属蛋白酶-1(MMP-1)及其抑制物-1(TIMP-1)的表达。结果与空白组相比,模型组软骨中MMP-1明显升高,TIMP-1降低,呈阴性表达,差异有统计学意义(P<0.05);与模型组和扶他林组相比,中药熏洗加电针组MMP-1明显降低,TIMP-1升高,差异有统计学意义(P<0.05)。结论中药熏洗加电针能有效治疗膝骨性关节炎,其作用机制之一可能与抑制MMP-1表达,升高TIMP-1表达有关。
Objective To explore the mechanism of steaming and washing with Chinese medicine combined acusector in treating articular genu osteoarthritis. Methods 32 rabbits were randomly divided into four groups: control group,model group, healing group and Dichofenac Diethylammon group. The anterior cruciate ligament was cut off to duplicate the articular genu osteoarthritis model. After that, the healing group was treated with Shangkexunxi decoction Combined acusector,once a day for four weeks. Dichofenac Diethylammon group was treated with Dichofenac Diethylammon for the same time. The expressions of MMP-1 and TIMP-1 were detected with immunohistochemistry. Results The expression of MMP-1 significantly increased in cartilage of model group com- pared with control group,while expression of TIMP-1 reduced(P〈0.05);the expression of MMP-1 significantly reduced in healing group compared with those in model group and Dichofenac Diethylammon group,while expression of TIMP-1 increased(P%0.05). Conclusion Steaming and washing with Chinese medicine Combined Acusector is effective in treating rabbit articular genu osteoarthritis,the partial mechanism is attributed to its inhibiting the expression of MMP-1 and increasing the expression of TIMP-1.
