摘要
以野生草莓(Fragaria vesca)EMC的匍匐茎尖为起始材料,对其茎尖培养及获得的组培苗的快繁和叶片再生进行了研究。结果表明,适合EMC茎尖培养的培养基为MS+0.5 mg/L6-BA+0.2 mg/L GA;适合组培苗快繁的培养基为MS+1.0 mg/L6-BA+0.2 mg/L GA+0.02 mg/L IBA;将EMC草莓叶片近轴面向下接入MS+2.0 mg/L TDZ+1.0 mg/L2,4-D培养基中,暗培养20 d后,愈伤组织形成率、不定芽再生率可分别达到100%和73%。本试验初步建立了一个有效的野生草莓EMC茎尖培养、组培苗快繁及叶片再生的体系。
The shoot tip culture, rapid propagation and leave regeneration of the wild strawberry EMC with shoot tip as initial material were studied out in this paper. The results showed that the adequate medium for EMC shoot tip culture was MS + 0. 5 mg/L 6 - BA + 0. 2 mg/L GA ; for rapid propagation of the tissue culture plantlets, the adequate medium was MS + 1.0 mg/L 6 - BA + 0. 2 mg/L GA + 0. 02 mg/L IBA ; In the medium of MS + 2.0 mg/L TDZ + 1.0 mg/L 2.4 - D , after 20 days dark treatment of the leaves with its near -axis surfaces in medium, the percentage of callus formation and the rate of its regeneration could reach 100% and 73%. In this study, an effective system of EMC shoot tip cultivation, multiplication and leaf regeneration was established.
