肝星状细胞-T6细胞系生长动态及分泌尿激酶型纤溶酶原激活物规律的研究 被引量:1

摘要 目的:观察肝星状细胞-T6(HSC-T6)细胞系的生长规律和动态,检测HSC-T6细胞上清液尿激酶型纤溶酶原激活物(uPA)抗原,探索HSC-T6分泌uPA的规律。方法:高糖DMEM培养液培养细胞,ELISA法测定培养细胞上清uPA含量,MTT法测定细胞的光密度值。结果:细胞生长高峰在第4天,2.0×10^5个/ml为最佳细胞接种浓度。细胞浓度在2.0×10^5-4.0×10^5个/ml时分泌uPA的能力较强,上清中uPA含量接近并明显高于其它组(P〈0.01)。随细胞浓度的增大,各浓度组HSC-T6细胞分泌uPA的含量呈明显增多。结论:HSC-T6细胞确能够合成和分泌uPA,其分泌含量有一定规律,呈明显细胞浓度依赖,第4-6天为细胞分泌高峰。 Objective: To observe the growth movement of hepatic-stellate-cell-T6 cell line, testing the amount of uPA excreted by HSC-T6 and secreting law. Methods : The cell was culture with high concentration of glucose and DMEM, the amount of uPA excreted was tested by HSC-T6 by way of ELISA, the optical density values of cell was tested by way of MTT. Results : The content of uPA has close correlation to the cell concentration and its growth time, which was in such order that its highest proliferation is on the 4th. Therefore 2.0 × 10^5/ml was the proper concentration for cell proliferation and the index could be tested. When concentration in 2.0 -4. 0× 10^5/ml, it secreted much more uPA. The content of uPA was almost the same and much higher (P 〉 0.05) than the other three groups (P 〈 0.01 ) . The more the concentration was, the more uPA were sereted. Conclusion: Make it sure for the first time that HSC-T6 actually can synthesize and secret uPA. The content of uPA has close correlation, the highest proliferation is on the 4th.
