摘要
目的建立测定清解合剂中大黄素、大黄酚含量的反相高效液相色谱(RP-H PLC)法。方法色谱柱为Krom asilC18柱(250m m×4.6m m,5μm),流动相为甲醇-磷酸盐缓冲液(75∶25),流速为1.0m L/m in,检测波长为254nm,柱温为45℃。结果大黄素与大黄酚进样量分别在21.12~105.6ng和29.28~146.4ng范围内与峰面积积分值线性关系良好,平均加样回收率分别为98.13%和98.43%,RSD分别为0.87%和0.71%(n=6)。结论RP-H PLC法专属性强、灵敏度高、干扰小、结果可靠,可用于清解合剂的质量控制。
Objective To establish a RP- HPLC method for the determination of the content of emodin and chrysophanol in Qinjie Mixture. Methods The samples were separated on Kromasil C18 column (250 mm × 4. 6 mm, 5 μm). The mobile phase consisted of methanol - phosphate buffer solution (75:25) at the flow rate of 1.0 mL/min. Tbe detection wavelength was 254 nm, and the column temperature was 45 ℃. Results The linear range of emodin was from 21.12 to 105.6 ng (r =0. 999 8). The average recovery rate was 98. 13% (RSD =0.87%, n=6).The linear range of chrysophanol was from 29. 28 to 146.4 ng (r=0.999 7). The average recovery rate was 98.43% (RSD= 0. 71%, n = 6). Conclusion The method is specific, sensitive and reliable in results with little interference. It can be used to the quality control for Qinjie Mixture.
