摘要
目的:观察氟伐他汀(Flu)对转化生长因子β_1(TGF-β_1)诱导下的大鼠肾小球系膜细胞(MCs)增殖和Ⅳ型胶原(ColⅣ)表达的影响,探讨其肾脏保护作用的可能机制。方法:选择处于对数生长期的MCs,分成对照组,TGF-β_1组,TGF-β_1加不同浓度Flu诱导组,采用MTT法检测MCs增殖,ELISA法检测ColⅣ表达。结果:在TGF-β_1诱导下,MCs的增殖随着Flu浓度的增加而逐渐下降,1μmol/LFlu即可明显抑制MCs增殖,P<0.01。TGF-β_1诱导下的MCs ColⅣ的表达随着Flu浓度的增加而逐渐下降,0.1μmol/L Flu即能明显抑制ColⅣ蛋白的表达,P<0.01。结论:氟伐他汀可呈剂量依赖性地抑制TGF-β_1诱导下MCs的增殖及MCs ColⅣ蛋白的表达,从而发挥其肾脏保护作用。
Objective:To investigate the effects of fluvastatin (Flu) on the cellular proliferation and the expression of type Ⅳ collagen ( ColⅣ ) in rat mesangial cells (MCs) induced by transforming growth factor-β1 (TGF-β1) ,and to explore the possible mechanisms of Flu for renoprotection. Methods:MCs were divided into the following groups according to different factors:Control group, TGF-β1 group, TGF-β1+ Fluvastatin (different concentration) group. The cellular proliferation was detected by MTT. The secretion of ColⅣ protein was quantitated by ELISA. Results:Flu could inhibit the proliferation of MCs induced by 5ng/ml TGF-β1 in a dose-dependent manner, 1μmol/L Flu could just suppress the proliferation significantly,P 〈0. 01. Flu could inhibit the expression of Col Ⅳ in MCs induced by 5ng/ml TGF-β1 in a dose-dependent manner :0. 1 μmol/L Flu could just suppress the expression of ColⅣ significantly, P 〈 0. 01. Conclusions:Fluvastatin can inhibit the cellular proliferation and the high expression of ColⅣ induced by TGF-β1 in a dose-dependent manner.
