摘要
确定了Gin活性肽的水解用酶为胰蛋白酶和碱性蛋白酶以及酶解过程中最佳酶解条件。通过两次酶解工艺制备Gln活性肽,胰酶的选用条件为底物浓度3%、酶浓度3%、pH8.0、温度48℃和反应时间2h;碱性蛋白酶的条件为底物浓度3%、酶浓度3%、pH8.0、温度68℃和反应时间1.5h。酶解液通过透析(10ku以下)后产品有效Gin为213.5mg/g。产物的分子质量在900~2.4×10^4u之间。
Trypsinase and alkaline proteinase were selected for Gin active peptide preparation, and the enzymolysis conditions were optimized. Result: The conditions for the trypsinase hydrolization are substrate concentration 3 %, enzyme concentration 3 %, pH 8.0, temperature 48℃, and reaction time 2 h; while the conditions for the alkaline proteinase hydrolization are substrate concentration 3%, enzyme concentration 3 %, pH 8.0, temperature 68℃, and reaction time 1.5 h. The concentration of effective Gin of the enzymolysis solution after dialysis (below 10 000 u) is 213.5 mg/g. The molecular weight of the product is between 900 u and 24 000 u.
