硒化复合L-氨基酸合成的扩大实验及产品质量检测

摘要 以豆粕为原料,利用酶水解工艺制备复合L-氨基酸,将复合L-氨基酸与亚硒酸钠螯合制备成硒化复合L-氨基酸,并在实验条件基础上进行扩大实验探索。试验得出豆粕酶解的最佳工艺条件:固液比为1∶25,pH为8.5,加酶量为10%胰蛋白酶加8%木瓜蛋白酶,酶解时间为4h加4h,豆粕水解率为39.61%.复合L-氨基酸与亚硒酸钠螯合的最佳工艺条件:温度为80℃,pH为9.0,摩尔配比为2∶1,时间为2h,螯合率为43.28%,扩大实验螯合率为37.96%,产品得率为52.99%.经红外光谱分析,复合L-氨基酸与亚硒酸钠之间存在螯合作用. Selenium compound-L-amino acids were prepared by chelating of sodium selenite and complex L-amino acid which was prepared by enzymatic hydrolysis from the material-soybean meal. Expanded experiment study was undertaken based on experiment conditions. The optimum enzymatic hydrolysis condition for complex L-amino acid was identified as follows., ratio of material to liquid as 1 : 25(W/V), pH 8.5, 10% of trypsin concentration for 4 h and 8% of papain concentration for 4 h another, the hydrolysis rate was up to 39.61%. The optimum chelating condition for sodium selenite and complex L-amino acid was 80% of chelating temperature, pH 9.0, the molar ratio was 2 : 1, chelating time was 2 h, then the chelating ratio reached 43.28%, the chelating ratio of expanded experiment was 37.96%, the product rate was 52.99%. The chelating of complex L-amino acid with sodium selenite was directly detected by infrared spectroscopy.
