摘要
用MS附加NAA、BA、KT等植物激素配制组合培养基,对诺丹冰草成熟胚进行了离体培养。结果表明,MS+NAA0.5mg/L+BA1.0mg/L和MS+NAA0.25mg/L+BA2.0mg/L是诺丹冰草胚诱导不定芽的两个理想组合培养基,对不定芽的形成和幼苗的生长有明显的促进作用,不定芽的诱导率分别达到64.0%和56.0%;显微切片观察到:不定芽起源于胚轴的表皮和皮层细胞,形成过程可分为启动阶段、芽原基形成阶段、叶原基形成阶段和不定芽形态建成阶段。
Mature embryos of Agropyron desertorum cv. ‘Nordan’ were cultured in MS based cultural medium with different concentrations of NAA, BA, or KT. Induced buds were sliced for microscopic observation. The results showed that MS based medium with the addition of NAA (0.25~0.5mg/L) or BA (1.0~2.0mg/L) had obvious promoting effects on induction of adventitious buds and growth of seedlings. It was observed that adventitious buds were formed in the epidermis and cortex cells of the embryonal axis. Formation of the adventitious buds could be divided into four stages, namely, initiation stage, bud primordium formation stage, leaf primordium formation stage and the stage of adventitious bud morphogenesis.
