免疫核糖核酸治疗羔羊痢疾的研究 Ⅰ.i-RNA的研制

用标准Ｂ型魏氏梭菌制备抗原免疫山羊，从免疫羊的肝、脾、淋巴结中按酚氯仿法稍加改进提取免疫核糖核酸（ｉＲＮＡ）。提取的ｉＲＮＡ制品用紫外分光光度计测定ＯＤ２６０ｎｍ／ＯＤ２８０ｎｍ＞２，ＯＤ２６０ｎｍ／ＯＤ２３０ｎｍ＞２，二苯胺法测定ＤＮＡ含量小于５％，Ｆｏｌｉｎ酚法测定蛋白质含量小于４％，据ＯＤ２６０ｎｍ值，计算出ｉＲＮＡ提取量为每克湿组织３ｍｇ左右。提取的ｉＲＮＡ做毒性、热源及变态反应检定合格，溶于含１２５ｉｕ／ｍｌ的肝素生理盐水中，ｉＲＮＡ含量为１ｍｇ／ｍｌ，除菌分装，－２０℃冰箱保存效果较好。

The liver,spleen and lymph of the goat immunized with B type clostridium perfringens were taken out.From which i RNA were extracted with phenol choroform method and determined its contents with ultrariolet photometer.The results were OD 260 nm/OD 280nm>2,OD 280 nm/OD 230 nm>2 U,DNA content <5%,protein content<45.According to OD 260nm, 3 mg i RNA could been extracted from 1 g wet tissue (liver,spleen,lymph node).According to the test of toxicity,heat source and allergic reaction,the i RNA was qualified and then retained in heparin physiological saline and put in -20℃ refrigerator.
