鸡传染性支气管炎病毒D_(41) 株S1基因部分序列的测定

SEQUENCING OF PARTIAL S1 GENE OF AVIAN INFECTIOUS BRONCHITIS VIRUS STRAIN D41

通过反转录及变退火温度ＰＣＲ方法扩增出含先导序列的ＩＢＶ广东地方分离株Ｄ４１的Ｓ１基因ｃＤＮＡ，长度约１．７ｋｂ．利用双脱氧链终止法对其５′端测序，准确读到３４２个碱基，并确定了翻译起始密码子ＡＴＧ的位置．

?Avian Infectious Bronchitis Virus(IBV) strain D41 is an attenuated vaccine strain of an isolate from Guangdong province. In this study, the cDNA of the D41 S1 gene (containing leader sequence, 1.7 kb) was amplified by reverse transcription(RT) and polymerase chain reaction(PCR), with alternating annealing temperature. Using dideoxy-mediated chain termination method, the sequencing assay results revealed the partial sequence of 5′ end of this segment(342bp) and the location of initiation codon ATG.
