表达H9亚型禽流感病毒HA基因的重组新城疫病毒的拯救 被引量:1

摘要 对现地分离的4株H9亚型禽流感病毒HA基因进行序列测定,选取其中1株在新城疫病毒LaSota弱毒疫苗株反向遗传操作系统的基础上,构建了表达H9亚型禽流感病毒野生型HA基因的重组新城疫病毒基因组eDNA克隆,经间接免疫荧光和RT—PCR鉴定,结果表明:拯救重组病毒为rL—H9HA:重组病毒MDT≥168h,ICPI和IVPI均为0,与亲本疫苗株LaSota具有相似的生长特性;重组病毒保持了LaSota弱毒疫苗亲本毒株对鸡胚良好的高滴度生长适应和低致病特性,具有作为同时预防H9亚型禽流感和新城疫的双价苗的应用前景。 In this study four strain H9 subtype avian influenza virus were isolated from commercial chickens, selected one strain by sequence analysis. On the base of reverse genetics of Lasota vaccine strain. To build the expression of H9 subtype of avian influenza virus in wildtype HA gene recombinant NDV genome eDNA cloning. The recombinant virus rL - H9 HA was confirmed by indirect immunofluoreseence assay and RT- PCR. The MDT of the recombinant virus is ≥168 h, furthermore both ICPI and IVPI is 0. The recombinant viruses maintained similar high growing titers in embryo and low pathogenicity as the parent strain did. Our results indicate that recombinant rL - H9HA virus can be used as a bivalent vaccine and have potential application in preventing H9 subtype avian influenza and ND.
