摘要
目的观察神经轴突导向因子Netrin-1在人肝细胞癌（HCC）组织中的表达及其对肝癌细胞侵袭能力的影响。方法应用免疫组织化学SP方法检测45例肝癌手术标本癌旁及癌组织中Netrin-1的表达水平，用卡方检验分析其与临床病理参数的关系。在肝癌细胞SMMC-7721中转染Netrin-1基因，观测过表达Netrin-1肝癌细胞侵袭能力的改变。结果Netrin-1在肝癌中的阳性表达率约为66．7％，主要在肝癌细胞胞质中表达，Netrin-1的阳性表达与性别、年龄、肿瘤大小以及细胞分化程度无关（P〉0．05），而与门静脉癌栓以及淋巴转移有关（P〈0．05）。细胞侵袭实验结果显示，与实验对照组比较过表达Netrin-1显著促进肝癌细胞SMMC-7721侵袭能力（P〈0．05）。结论HCC组织中高表达的Netrin-1与肿瘤侵袭转移有关，Netrin-1促进肝癌细胞侵袭能力可能是肝癌转移机制之一。
Objective To study Netrin-1 expression in hepatocellular carcinoma tissue and its effect on invasion of hepatoeellular carcinoma cells. Methods The expression of Netrin-1 in 45 cases of hepatocellular carcinoma was detected by immunohistoehemistry. The correlation between Netrin-1 and clinieopathologic features was analyzed by Chi-Squared test. The effect of Netfin-1 on invasion of SMMC- 7721 cells was analyzed by invasion assay. Results The positive rate of Netrin-1 in HCC tissue was 66.7%. Netrin-1 expression was mainly located in kytoplasm of liver cancer cells. Netrin-1 expression was associated with lymph node metastasis, portal vein invasion, and metastatic lesions （ P 〈 0.05 ）, bot not with sex, age, tumor size, and cell differentiation （ P 〉 0.05 ）. Cell invasion assay revealed that the number of migrating SMMC-7721 cells induced by Netfin-1 was increased （ P 〈 0.05 ）. Conclusion The up-regu- lation of Netrin-1 in HCC has a close relationship to the invasion of hepatoma cells. Invasion of hepatoma cells induced by Netrin-1 may be one of mechanisms of metastasis of HCC.
