摘要
设计了能同时检测Re MV、TMV和ToMV的简并引物,利用RT-PCR的方法对我国不同地区的地黄病毒样品进行了检测。共克隆了25个病毒样品的外壳蛋白(CP)基因,序列测定结果表明,24个病毒样品均是Re MV,其相近病毒TMV未检测到,ToMV仅在一个野生地黄样品上检出,说明Re MV在我国栽培地黄和野生地黄上普遍存在。
Virus somples infected Rehmannia glutinosa in different regions were detected by using RT--PCR with the degenerate primers designed to detect Rehmannia mosaic virus (ReMV),Tobacco mosaic virus (TMV) and Tomato mosaic virus (ToMV)simuhaneously. Coat protein (CP) genes of 25 virus samples were cloned and sequenced. The results indicate that the 24 virus samples were ReMV ,TMV was not detected,the ToMV is detected only in one wild Rehmannia glutinosa sample. According to the above experiment,ReMV is the major virus in cultural and wild Rehmannia glutinosa in China.
