摘要
采用海湾红宝石李(PrunussalicinaLindl.cv.Gulf-ruby)嫩梢茎段为外植体,研究了初代培养过程中MS、改良MS(1/2 NH4NO3)、3/4 MS、WPM基本培养基以及生根培养过程中1/2 MS、1/2改良MS、1/2 WPM、WPM对其组织培养过程中的增殖和生根的影响。结果表明,在初代培养过程中,以WPM为基本培养基效果最好,60d后增殖系数(4.07)和1cm以上嫩茎比例(28.2%)均最高;改良MS培养基效果次之;MS和3/4 MS培养基没有区别,培养效果最差。在生根培养过程中,1/2 MS培养基和WPM培养基效果较好,生根率分别为10.4%和10.1%,生根倍数均为2.0。
The effects of four basal media, MS,improved MS, 3/4 MS and WPM on multiplication and the other four basal media, 1/2 MS, 1/2 improved MS, 1/2 WPM and WPM on the root induction of Gulf-ruby plum (Prunus salicina Lindl. cv. Gulf-ruby) were studied. During initial culture, using WPM as the basal medium obtained the highest multiplication rate(4. 07) and the highest rate (28. 02%) of tender shoot above 1 cm height culturing for 60 days; while using improved MS, the lower;and the lowest for MS and 3/4 MS. The shoot rooting rates were 10.4% and 10. 1% for 1/2 MS and WPM,respectively,and the rooting times for the two media were the same,2.0.
