替米沙坦对大鼠心肌缺血再灌注损伤的保护作用 被引量:4

摘要 目的探讨替米沙坦对大鼠心肌缺血再灌注损伤(MIRI)的保护作用及机制。方法将30只雄性Wistar大鼠随机分为假手术组(A组)、缺血再灌组(B组)、缺血再灌注加替米沙坦组(C组),每组10只。B组与C组大鼠在体结扎冠状动脉前降支30min后,松扎再灌注120min,制备心肌缺血再灌注模型。A组及B组于术前24h及2h灌胃生理盐水5ml/kg,C组术前24h及2h灌胃替米沙坦10mg/kg(以生理盐水配制)。生化自动分析仪测定血清天门冬氨酸氨基转换酶(AST)、心肌型肌酸激酶同功酶(CK-MB)及乳酸脱氢酶(LDH)活性,比色法测定血清丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,放免法测定血浆内皮素(ET)、血栓素A2(TXA2)及前列环素(PGI2)含量。NBT染色计算心肌梗死面积(MIS),光镜和电镜观察心肌组织形态学。结果与B组比较,C组能明显缩小MIS(P<0.01),降低血清AST、LDH、CK-MB(P<0.05或<0.01)及MDA活性(P<0.01),提高SOD、GSH-Px活性(P<0.05),降低血浆TXA2含量,提高PGI2水平及PGI2/TXA2比值(P<0.05或<0.01),并能明显减轻心肌组织水肿以及超微结构的损伤。结论替米沙坦对大鼠心肌缺血再灌注损伤具有明显保护作用,可能与其抗自由基损伤,增强抗氧化酶活性,抑制脂质过氧化损伤,扩张血管,调节PGI2/TXA2平衡等途径发挥抗心肌缺血再灌注损伤作用。
