原发性高血压患者外周血单核细胞趋化因子-1基因表达及意义

摘要 目的:观察原发性高血压(EH)患者外周血单核细胞趋化因-1(MCP-1)基因表达,探讨其与EH的关系及意义。方法:用酶联免疫吸附(ELISA)法测定62例EH患者和30例正常对照者外周血MCP-1蛋白含量,用逆转录聚合酶链反应(RT-PCR)法检测外周血单个核细胞中MCP-1 mRNA表达。结果:与对照组比较,EH组外周血MCP-1蛋白含量及外周血单个核细胞MCP-1 mRNA表达均显著增高(P<0.01)。结论:(1)MCP-1可能参与了EH的病理生理过程;(2)外周血单个核细胞的MCP-1 mRNA表达上调可能是EH患者外周血MCP-1蛋白含量增高的机制之一。 Objective: To observe the level changes of protein and mRNA expression of monocyte chemotactic protein-1(MCP-1) in peripherial blood of patients with essential hypertension(EH),and to probe the relationship between MCP-1 and EH.Methods: Concentrations of circulating MCP-1 of 62 EH patients(EH group) and 30 normotensive subjects(control group) were measured with enzyme linked immunosorbent assay(ELISA),and the expression of MCP-1 mRNA in mononuclear cells were measured with reverse transcription polymerase chain reaction.Results: Protein MCP-1 in peripherial blood and expression of mRNA MCP-1 in mononuclearcell in peripherial blood of EH group were significantly higher than those of control group(P〈0.01).Conclusions: MCP-1 in peripherial blood might participate in the patho-physiology process of EH.The up-regulating expression of MCP-1 mRNA may be one of the mechanisms that increase protein MCP-1 level in peripherial blood of EH patients.
