摘要 目的:探讨用免疫磁珠分离法(magnetic activated cell sorting,MACS)从小鼠股骨、胫骨分离纯化骨髓造血干细胞CD117的方法。方法:收集小鼠骨髓细胞,台盼蓝染色观察其活力,用免疫磁珠分离法分选CD117细胞,流式细胞仪检测荧光标记CD117表达阳性率。结果;未分选前CD117细胞纯度为(41.56±18.77)%,MACS分选CD117细胞纯度(88.68±4.74)%,纯化前后有明显差异(P<0.05)。结论:MACS阳性选择法分选系统能有效分选骨髓造血干细胞CD117。 Objective: To develop a method for purification of CD117 cells from murine bone marrow using magnetic activated cell sorting(MACS) method with immunomagnetic beads.Methods: Bone marrow cells were obtained from mice thighbone and tibia.CD117+ cells were isolated from these cells using immunomagnetic beads on MiniMACS device,and hematopoietic stem cells(HSCs)were purified.Viability of the collected cells was determined with flow cytometry and light microscopy,respectively.Results: The purity quotient of CD117 HSCs increased from(41.56±18.77)% to(88.68±4.74)% through MACS,and the difference was significant(P〈0.05).Conclusion: Highly purified CD117 cells can be obtained from murine bone marrow by positive selection using MACS withont any inpairment to the viability of the derived cells.
