摘要
[目的]探讨五鹤续断愈伤组织的培养及增殖。[方法]以五鹤续断初春萌发的幼嫩茎、叶为外植体,研究不同植物生长调节物质及其组合对五鹤续断愈伤组织的诱导效应。[结果]当NAA浓度为1.0 mg/L时,对五鹤续断愈伤组织的诱导率最高,为65.4%。当2,4-D浓度为1.0 mg/L时,对五鹤续断愈伤组织的诱导率最高,为60.2%。当6-BA浓度为1.5 mg/L时,对五鹤续断愈伤组织的诱导率最高,为67.2%,且其诱导的愈伤组织的生长势比NAA的更好。当KT浓度为0.5 mg/L时,对五鹤续断愈伤组织的诱导率最高,仅为35.2%。6-BA 1.5 mg/L+NAA 0.2 mg/L和6-BA 1.0 mg/L+2,4-D 2.0 mg/L对五鹤续断愈伤组织的诱导率分别达89.5%和92.7%。在黑暗和光照培养条件下,愈伤组织的生长曲线均呈"S"型,符合Logistic生长模型。[结论]该研究为五鹤续断次生代谢物的提取和多倍体诱导提供了理论依据。
[Objective] The purpose was to discuss the culture and proliferation of Wuhe dipsacus callus. [Method] With tender stems and leaves germinated in early spring of Wuhe dipsacus as explants, the induction effects of different plant growth regulators and their combinations on Wuhe dipsacus callus were researched. [Result] When the conch, of NAA was 1.0 mg/L, its induction rate on Wuhe dipsaeus callus was highest, being 65.4%. When the conch, of 2,4-D was 1.0 mg/L, its induction rate on Wuhe dipsacus callus was highest, being 60.2%. When the conch, of 6-BA was 1.5 mg/L, its induction rate on Wuhe dipsacus callus was highest, being 67.2% and the growth potential of Wuhe dipsacus callus induced with 6-BA was better than that of Wuhe dipsacus callus induced with NAA. When the conch, of KT was 0.5 mg/L, its induction rate on Wuhe dipsacus callus was highest, being only 35.2%. The induction rates on Wuhe dipsacus callus of 6-BA 1.5 mg/L + NAA 0.2 mg/L and 6-BA 1.0 mg/L + 2,4-D 2.0 mg/L were up to 89.5 % and 92.7% resp. The growth curve of calli culturnd in darkness and daylight showed to "S" type and accorded with Logistic growth model. [Conclusion] The research provided theoretical foundation for the extraction of secondary metabolites in Wuhe dipsacus and its polyploid induction
