摘要
[目的]研究纤维素酶对大豆秸秆、豆皮的最优水解条件和大豆秸秆的最佳预处理条件,为提高秸秆糖化效果及饲料转化率提供参考。[方法]以大豆秸秆、豆皮为材料,研究纤维素酶水解大豆秸秆的最适温度、pH值、底物浓度和大豆秸秆的最佳预处理条件。[结果]纤维素酶水解大豆秸秆的最适酶解温度为40℃,最适酶解pH值为6。在最适温度和pH值条件下酶的催化活性可维持9~10h。底物经表面活性剂吐温.20和吐温-80处理后可以将产糖量提高,在两种处理条件下,糖产量分别可提高28%(吐温-20)和38%(吐温-80)。经薄层层析,水解液中含有葡萄糖和麦芽糖两种成分。[结论]纤维素酶水解大豆秸秆、豆皮的最优条件为40℃、pH值6,并且表面活性剂吐温-20和吐温-80可不同程度地提高大豆秸秆产糖量。
[Objective] The best condition of hydrolysis of soybe.an straw and seed skin, and the optimal pretreatment condition of soybean straw were studied for the reference for efficiency-raising of the saccharify of straw and feed-using. [Method] The soybean straw and seed skin being taken as the experimental material, the optimum temperature, pH value, the concentration of substrate in the process of the hydrolysis, and the optimum pretreatment of soybean straw were studied. [ Results ] The results showed that the optimum temperature was 40 ℃ and the pH value, 6. In the optimum temperature and pH conditions, the activity of cellulose could be maintained for 9 - 10 hours. When the straw powder be treated by the surfactant Tween -20 and Tween - 80,28% mad 38% of the sugar yield could be increased. After the thin layer chromatography, there were two components of glucose and maltose in the hydmlyzate. [Conclusion] Cellulase hydrolysis of soybean straw, Yuba The optimal condition was as follows: the temperature of 40 ℃, pH value 6, and the surfactant Tween -20 and Tween -80 could improve sugar yield of soybean straw to different extent.
